Restriction digests of the clone give the following sizes (kb): HindIII/KpnI--2.7, 2.1; PvuII--2.4, 1.3, 0.96; SacI--3.5; 1.3. Insert contains the following restriction sites (approximate kb from the 5' end): PvuII--1.13, 1.19; SacI--0.79. The plasmid affects the growth of transformed bacteria and viability in stationary phase. Growth at 30C reduces product degradation. The following primers can be used to amplify a 0.678 kb pspA product from the insert or genomic DNA (nt 1312-1990 of the published sequence): LSM1 5'-CCGGATCCAGCTCCTGCACCAAAAC-3'; LSM2 5'-GCGCTGCGACGGCTTAAACCCATTCACCATTGG-3'. |
